چکیده

Rare events are often described by a Poisson distribution. In clinical research the examination of such rare events could be the basis of a non-inferiority trial. In order to plan such a trial the power of a statistical test comparing two Poisson distributions is required. The purpose of this paper is to present a method for calculating size and power of three statistical tests. The method can be easily realized with a short SAS R © program. The paper will depict the approach graphically and theoretically.

Sample Size Estimation for Non-Inferiority Trials: Frequentist Approach versus Decision Theory Approach

BACKGROUND Non-inferiority trials are performed when the main therapeutic effect of the new therapy is expected to be not unacceptably worse than that of the standard therapy, and the new therapy is expected to have advantages over the standard therapy in costs or other (health) consequences. These advantages however are not included in the classic frequentist approach of sample size calculatio...

Sample Size Formulas for Non-Inferiority Clinical Trials with Time-to-Event Data

In many pharmaceutical studies, non-inferiority clinical trials are usually conducted because of the difficulty of finding a therapy that has more superior efficacy than a recognized effective one. When planning the non-inferiority clinical trials with a time-to-event endpoint, the calculation of sample size is one of the most fundamental steps. A proper sample size provides reasonable power to...

Do non-inferiority trials of reduced intensity therapies show reduced effects? A descriptive analysis

OBJECTIVES To identify non-inferiority trials within a cohort where the experimental therapy is the same as the active control comparator but at a reduced intensity and determine if these non-inferiority trials of reduced intensity therapies have less favourable results than other non-inferiority trials in the cohort. Such a finding would provide suggestive evidence of biocreep in these trials....

Application of Gompertz-Poisson Distribution in LifetimeTheory

Gompertz-Poisson distribution is a three-parameter lifetime distribution with increasing, decreasing, increasing-decreasing and unimodal shape failure rate function and a composition of Gompertz and Poisson distributions cut at zero point that in this paper estimated the parameters of the distribution by maximum likelihood method and in order to confirm the calculated estimates, based on random...

Retrospective Assessment of Non-Inferiority in the Rare Disease, Guillain–Barre Syndrome

After a non-inferiority margin is established, a prospective non-inferiority trial is usually conducted to confirm the noninferiority of the new product when compared to the existing product. Non-inferiority trials typically require considerably larger sample sizes than placebo-controlled trials [4]. This is due to the fact that the margin of equivalence (non-inferiority) is often much smaller ...
